Obesity-Activated Lung Stromal Cells Promote Myeloid Lineage Cell Accumulation and Breast Cancer Metastasis

چکیده

Obesity is correlated with increased incidence of breast cancer metastasis; however, the mechanisms underlying how obesity promotes metastasis are unclear. In a diet-induced obese mouse model, enhanced lung in both presence and absence primary mammary tumors recruitment myeloid lineage cells into lungs. tumors, mice demonstrated numbers elevated collagen fibers within stroma, reminiscent premetastatic niches formed by tumors. Lung stromal isolated from tumor-naïve showed proliferation, contractility, expression extracellular matrix, inflammatory markers transforming growth factor beta-1 (TGFβ1). Conditioned media promoted cell migration vitro response to colony-stimulating 2 (CSF2) invasion tumor cells. Together, these results suggest that prior formation, alters microenvironment, creating conducive metastatic growth.

Atorvastatin Sensitizes Breast and Lung Cancer Cells to Ionizing Radiation

Tumour cells may be resistant to radiotherapy that results in unsuccessful cancer treatment in patients. The aim of this study was to evaluate the sensitizing effect of atorvastatin (ATV) on breast cancer (MDA-MB-231) and non-small cell lung cancer (A-549) cells following exposure to ionizing radiation (IR). These cells were treated with ATV and exposed to X-ray at dose 4 Gy. The radiosensitizi...

Tissue-resident stem cells promote breast cancer growth and metastasis.

Mesenchymal stem cells derived from bone marrow have recently been described to localize to breast carcinomas and to integrate into the tumor-associated stroma. In the present study, we investigated whether adipose tissue-derived stem cells (ASCs) could play a role in tumor growth and invasion. Compared with bone marrow-derived cells, ASCs as tissue-resident stem cells are locally adjacent to b...

Osteopontin promotes CCL5-mesenchymal stromal cell-mediated breast cancer metastasis.

The interaction between cancer and its local microenvironment can determine properties of growth and metastasis. A critical component of the tumor microenvironment in this context is the cancer-associated fibroblast (CAF), which can promote tumor growth, angiogenesis and metastasis. It has been hypothesized that CAF may be derived from mesenchymal stromal cells (MSC), derived from local or dist...

Geminin facilitates FoxO3 deacetylation to promote breast cancer cell metastasis.

Geminin expression is essential for embryonic development and the maintenance of chromosomal integrity. In spite of this protective role, geminin is also frequently overexpressed in human cancers and the molecular mechanisms underlying its role in tumor progression remain unclear. The histone deacetylase HDAC3 modulates transcription factors to activate or suppress transcription. Little is know...
